目的：本次实验以主要人肺腺癌A549细胞为研究对象,以此来认识白藜芦醇能够诱导肺癌A549细胞凋亡.方法：通过倒置显微镜、荧光显微镜观察不同浓度白藜芦醇作用于A549细胞在24h、48h和72h的细胞生长形态,同时,利用台盼蓝拒染法检测白藜芦醇对A549细胞的生长抑制率,进而确定IC50值,以及细胞的凋亡情况,再通过流式细胞术检测细胞周期和细胞线粒体膜电位的变化.结果证实白藜芦醇作用在人肺癌A549细胞48h时呈现出不同程度的凋亡性状,同时显示量效关系.白藜芦醇诱导A549细胞凋亡作用在48h的最佳浓度分别为110µg/mL、80µg/mL,且IC50值分别为108.002µg/mL、77.462µg/mL.最后经过流式细胞术检测结果可知,白藜芦醇可将A549细胞滞停于S期和G2期,同时线粒体膜电位伴随浓度的增加而逐渐降低.结论：白藜芦醇诱导人肺腺癌A549细胞凋亡,其IC50值分别为108.002μg/mL、77.462μg/mL.线粒体膜电位随浓度增加而降低.并且细胞阻滞于G2期和S期,进而诱导细胞凋亡.
